Who Is This For?

This tool is built for:

Pharmaceutical scientists comparing pre- and post-change dissolution profiles

QC and QA analysts performing dissolution profile comparisons for regulatory submissions

Anyone who needs accurate, audit-ready f1 and f2 results without building formulas from scratch

If you’ve ever spent 20 minutes double-checking a formula in a plain Excel sheet — or worried you had the wrong parentheses in your f2 calculation — this tool was built for you.

Table comparing dissolution profiles for prechange and postchange samples with statistical analysis and legend explanations.
Table listing features with their descriptions, including automatic f1 & f2 calculation, full-precision means, 85% rule check, CV threshold check, and pass/fail output.

Everything is handled inside Excel — no internet connection required, no data uploaded anywhere, no new software to learn.

How to Use It

Step 1:  Enter your time points

Input the nominal time points for your dissolution study (e.g., 5, 10, 15, 30, 45, 60 min).

Step 2:  Enter your raw replicate values

Input the individual dissolution values for each unit at each time point — up to 12 replicates per time point. The calculator computes the means at full precision automatically.

Step 3:  Review the automated checks

The calculator flags any time points that violate the 85% rule or exceed CV thresholds before the calculation runs.

Step 4:  Read your f1 and f2 results

The results summary shows your calculated f1 and f2 values with clear Pass/Fail indicators against acceptance criteria.

Regulatory Basis

The f1 and f2 calculations in this tool follow the methodology described in:
✓  FDA Guidance for Industry: Dissolution Testing of Immediate Release Solid Oral Dosage Forms (1997)
✓  EMA Guideline on the Investigation of Bioequivalence (CPMP/EWP/QWP/1401/98 Rev. 1)
Acceptance criteria applied: f1 ≤ 15 and f2 ≥ 50.

The Problem with Manual f1/f2 Calculations

Building f1 and f2 in a plain Excel sheet looks straightforward. In practice, there are at least four ways to get the wrong answer — silently.

•       Misplaced parentheses that apply the −0.5 exponent to the wrong expression

•       Forgetting to exclude time points where Rt > 85% (the 85% rule)

•       Rounding means before using them in the calculation

•       No CV check, so non-qualifying data gets included without warning

The troubling part: Excel produces a number either way. You won’t always know when something is wrong.
